Get things ready

You will need these items for the protocol.

Hardware

• BactoBox® setup including a flow cell.

  • HW: 7.6a

  • SW: v2026.02a

• Incubator shaker

• Vortex mixer

• Rack for holding 5 and 15 mL centrifuge vials.

• Cold rack for holding 5 and 15 mL centrifuge vials, e.g. ice rack, chillblock, or mini cooler. You can also use a simple expanded polystyrene with a mix of water and ice (or a cooling element).

• Empty 2, 5, or 15 mL centrifuge vials for sampling.

• Serological pipette, preferably 2 mL version

• Serological pipette controller

• P200 pipette.

• Sterile pipette tips.

Reagents

• 1 × culture in deceleration phase. Preferably use a bioreactor culture. Alternatively, you can also use a 50 mL shake flask culture in 250 mL vessel to simulate the process.

• Dilution vials either ready-to-use or freshly prepared using the dilution dispenser.

• Disinfection vials.
