Qualification
Qualification happens in two phases:
Phase A: Measure with a QC vial (Phase A: QC vial)
Phase B: Measure with a dilution vial (Phase B: Dilution vial)
In phase A, BactoBox® both tunes and qualifies. This happens simultaneously.
In phase B, we qualify further.
The setup passes qualification if it passes both phase A and phase B. When we say setup we mean the combination of all items: BactoBox®, flow cell, tubing kit, external filter, etc.
Always qualify on setup change
Qualify whenever you change the setup. That is, whenever you replace an item. Even if you just change the external filter.
Get things ready
Prepare 2 × dilution vials
Find two dilution vials and place them the vial rack next to the tubing kit. We'll use these dilution vials shortly (one for each phase).

Use SBT's dilution vial for the best results
SBT has a rigorous quality control procedure to ensure that the dilution vial pairs perfectly with BactoBox®.
Only use non-SBT dilution liquid if you absolutely must
Always use filtered, gamma-sterilized dilution liquid. In-house or third-party sterile-filtered (0.22 µm) dilution liquid is acceptable. However, make sure to filter the liquid immediately before use to avoid unwanted background particles.
That is it!
Next, we make a QC vial. See Make QC vial.
